What is Afanix?
Afanix 40mg is a kinase inhibitor indicated for the first-line treatment of patients with metastatic non-small cell lung cancer (NSCLC) whose tumors have an epidermal growth factor receptor (EGFR) deletion. exon 19 or exon 21 substitution mutation (L858R) detected by an FDA-approved Trial & Treatment for patients with advanced squamous, metastatic NSCLC following platinum-based chemotherapy.
Uses of Afanix
Afanix is used to treat:
- Non-small cell lung cancer (NSCLC) with mutations in the EGFR gene (deleting exon 19 and/or exon 21 mutation L858R);
- Non-small cell lung cancer (NSCLC) metastatic squamous cell carcinoma with mutations in the EGFR gene.
How Afanix works
Afatinib binds covalently to the kinase domains of EGFR (ErbB1), HER2 (ErbB2) and HER4.
(ErbB4) and irreversibly inhibit tyrosine kinase autophosphorylation, leading to downregulation of ErbB signaling.
Afatinib has been shown to inhibit autophosphorylation and proliferation in vitro of cell lines expressing wild-type EGFR or genes expressing EGFR exon 19 deletion mutations or selected exon 21 L858R mutations, including Including several secondary T790M mutations, concentrations of Afatinib were achieved, at least transiently, in patients. In addition, Afatinib inhibited in vitro the proliferation of HER2-overexpressing cell lines.
Afatinib treatment resulted in inhibition of tumor growth in transplanted nude mice that overexpress wild-type EGFR or HER2 or in the EGFR L858R/T790M dual mutant model.
What is the dose of Afanix?
Recommended dose: The recommended dose of Afatinib is 40 mg orally once daily until disease progression or no longer tolerated by the patient.
Your doctor may increase or decrease your dose depending on how well you tolerate Afanix
Missed dose of Afanix
- If you forget a dose, take it as soon as you remember, if it is almost time for your next dose, skip the missed dose.
- Do not miss two doses in a row and never take a double dose to make up for the missed dose.
- Call your doctor for instructions if you miss more than one dose.
Overdosage of Afanix
- Seek immediate medical attention or contact your doctor if an overdose of this medicine is suspected.
- Bring your prescription and medicine box to your doctor.
How to take Afanix
Take Afanix by mouth on an empty stomach at least 1 hour before or 2 hours after eating.
Take Afanix on your regular dosing schedule about 24 hours apart, unless otherwise directed by your doctor.
Keep taking Afanix medicine even if you feel well and do not miss any doses while you are using it.
Afanix side effects
Common side effects of Afanix include:
- Mild diarrhea for 1 day or less.
- Nausea, vomiting, loss of appetite.
- Mouth Sores.
- Acne, itching, dry skin.
- Redness, pain, swelling, or other signs of infection around your fingernails or toenails.
Safety tips when taking Afanix
Take Afanix 40 mg on an empty stomach at least 1 hour before a meal or 2 hours after a meal.
Diarrhea is common with Afanix 40 mg Tablet and can sometimes be severe. Your doctor may prescribe medication to treat diarrhea. Stay hydrated and take this medication exactly as directed.
Afanix 40 mg may make your skin sensitive to the sun. Limit your time in the sun. Use sunscreen and wear protective clothing if you must be exposed to the sun.
Afanix 40 mg may cause redness, rash and acne. Treat skin reactions as soon as you notice them.
Use a reliable method of birth control to prevent pregnancy while you are taking this medicine and for one month after you stop taking it.
Monitor your blood pressure regularly while taking this medicine. Notify your doctor if you notice symptoms of very high blood pressure such as severe headache, confusion, vision problems, nausea or vomiting.
It can cause serious bleeding problems. Notify your doctor if you have a headache, an upset stomach, or if you notice blood in your urine or stools.
Do not take this medicine if you are pregnant, planning to conceive or breast-feeding.
Afanix drug interactions
P-gp inhibitor administration (Ritonavir at 200 mg twice daily) 1 hour before Afatinib administration increased systemic exposure to Afatinib by 48%. There was no change in Afatinib exposure when Ritonavir was administered concomitantly with or 6 hours after Afatinib.
Co-administration of P-gp inhibitors (including but not limited to Ritonavir, Cyclosporine A, Ketoconazole, Itraconazole, Erythromycin, Verapamil, Quinidine, Tacrolimus, Nelfinavir, Saquinavir, and Amiodarone) may increase exposure to Afatinib. Afatinib.
Co-administration of an oral dose of a P-gp inducer (Rifampicin at 600 mg once daily for 7 days) reduced exposure to Afatinib by 34%.
Co-administration of P-gp inducers (including but not limited to Rifampicin, Carbamazepine, Phenytoin, Phenobarbital and St. John's wort) with Afatinib may reduce exposure to afatinib.
How to store Afanix?
- Store at controlled room temperature 15° – 30°C
- Protect this medication from light and moisture.
- Do not use the medicine beyond the expiry date indicated on the outside of the medicine box.
