What is Mekinist?

Mekinist 2mg (trametinib) is a cancer medicine that interferes with the growth and spread of cancer cells in the body.

Uses of Mekinist

Mekinist is used alone or in combination with another medicine called dabrafenib (Tafinlar) to treat certain types of cancer in people with a gene mutation.

Mekinist is used to treat:

  • Melanoma (skin cancer) that cannot be treated with surgery or has spread to other parts of the body, or to prevent melanoma from returning after surgery;
  • Non-small cell lung cancer that has spread to other parts of the body; or
  • Advanced or metastatic thyroid cancer has spread and there are no other treatment options. 

How does Mekinist work?

Trametinib belongs to a class of anti-cancer drugs known as anticancer drugs, and specifically to a class of drugs known as protein kinase inhibitors. It is used alone or with dabrafenib to treat a specific type of melanoma (skin cancer) that cannot be surgically removed or has metastasized (spread to other parts of the body). It is also used after surgery to remove melanoma and help prevent melanoma from coming back. Trametinib is also used in combination with dabrafenib to treat metastatic non-small cell lung cancer.

Trametinib is not suitable for all types of melanoma or non-small cell lung cancer. It will only interfere with the growth of skin cancer cells that have a specific genetic mutation, called BRAF. Your doctor will check for this mutation before prescribing trametinib.

What is the dose of Mekinist?

Usual Adult Dose for Melanoma – Metastasis:

  • Single agent: 2 mg orally once a day. 
  • Combination therapy: 2 mg orally once a day with dabrafenib 150 mg orally twice a day. 
  • Duration of treatment: Until disease progression or unacceptable toxicity occurs.

Missed dose of Mekinist

The missed dose should be taken as soon as possible. You should skip the missed dose if it is time for your next scheduled dose. Do not use extra medicine to make up for the missed dose.

Overdosage of Mekinist

  • Seek immediate medical attention or contact your doctor if an overdose of Mekinist is suspected.
  • Bring your Mekinist prescription and box to your doctor.

How to take Mekinist 

Take Mekinist exactly as prescribed by your doctor. Follow all directions on your medication label and read all medication instructions or instruction sheets. Your doctor may sometimes change your dose. Use the medication exactly as directed.

Mekinist side effects

The following adverse events were common (occurring in more than 30%) in patients receiving Mekinist:

  • Aspartate aminotransferase (AST) elevation
  • Rash
  • Diarrhea
  • Low albumin (Low albumin)
  • Increased Alanine aminotransferase (ALT)
  • Anemia
  • Lymphedema

These side effects are less common (occurring in about 10-29%) of patients taking Mekinist:

  • Increased alkaline phosphatase
  • Acne vulgaris (acne)
  • Stomatitis (stomatitis)
  • High Blood Pressure
  • Stomachache
  • Hemorrhage
  • Dry skin
  • Itchy
  • Paronychia (nail infection)

Precautions while using Mekinist

You should not use Mekinist if you are allergic to trametinib.

To make sure Mekinist is safe for you, tell your doctor if you have:

  • Heart disease , high blood pressure;
  • Diabetes;
  • Lung disease;
  • Liver or kidney disease;
  • Stomach or bowel disorders;
  • eye problems (especially problems with your retina); 
  • Bleeding problems or blood clots.

Using Mekinist with dabrafenib may increase your risk of developing a certain type of skin cancer. Ask your doctor about your specific risk. Tell your doctor if you notice any new symptoms on your skin such as redness, warts, sores that don't heal, or a mole that changes in size or color.

Trametinib may harm an unborn baby or cause birth defects if the mother or father is using this medicine.

If you are a woman, do not use Mekinist if you are pregnant. You may need to have a negative pregnancy test before starting this treatment. Use effective birth control to prevent pregnancy while you are using this medicine and for at least 4 months after your last dose.

If you're a man, use effective birth control if your partner can get pregnant, even if you've had a vasectomy. Continue using birth control for at least 4 months after your last dose

Tell your doctor right away if pregnancy occurs while mom or dad is using Mekinist.

This medicine may affect fertility (ability to have children) in women. However, it is important to use birth control to prevent pregnancy because trametinib can harm an unborn baby.

You should not breast-feed while using this medicine and for at least 4 months after your last dose.
