How does Gefitinib work?

The active ingredient gefitinib is a selective small-molecule inhibitor of the epidermal growth factor receptor tyrosine kinase and is an effective treatment for patients whose tumors have EGFR tyrosine kinase region-activating mutations regardless of any therapy.
Common EGFR-activating mutations have robust response data supporting sensitivity to gefitinib; for example, progression free heart rate (95% CI) was 0.489 (0.336, 0.710) for gefitinib compared with doublet chemotherapy.
Response data to gefitinib are sparser in patients with tumors harboring less common mutations; available data indicate that G719X, L861Q and S7681 are susceptible mutations; and T790M alone or exon 20 insertion are resistance mechanisms.
