What is Prograf 1mg Tacrolimus ?
Prograf contains the active ingredient Tacrolimus, which reduces the body's immune system. The immune system helps the body fight infections. The immune system can also fight or "reject" a transplanted organ such as a liver or kidney.
This happens because the immune system sees the newly transplanted organ as an invader. It is used together with other medicines to prevent your body from rejecting a heart, liver, or kidney transplant.
Mechanism of action of the active ingredient Tacrolimus
At the molecular level, the effects of Tacrolimus are mediated through binding to a cellular protein (FKBP12) responsible for the intracellular accumulation of the compound. The FKBP12-Tacrolimus complex specifically binds and competes with and inhibits calcineurin, resulting in inhibition of the calcium-dependent T-cell signaling pathway, thereby preventing transcription of a discrete lymphokine genome .
Tacrolimus is a potent immunosuppressant and has demonstrated experimental activity. In particular, the active substance inhibits the formation of cytotoxic lymphocytes, which are the main cause of transplant rejection.

Dosage of Prograf
Prograf injection: dilute with 0.9% NaCl or 5% dextrose to get concentrations of 0.004 mg/ml and 0.02 mg/ml before use; The first dose should be administered within the first 6 hours after organ transplantation, 0.03-0.05 mg/kg/day IV, administered with corticosteroids, continued until the patient can take tablets.
Prograf tablets: adult kidney transplant: start 0.2 mg/kg/day, adult liver transplant: start 0.10-0.15 mg/kg/day, pediatric liver transplant: 0.15-0.20 mg/kg/day, dose split every 12 hours.
